Alginates are polysaccharides which are naturally present in the cell wall of brown algae and extracted because of their rheological properties. The alginate industry uses an extraction protocol, which central step, the carbonatation, is conducted in batch reactors. It is a high water, reagent and time demanding process. The main objective of this thesis is to optimize the alginate extraction process. A study of the existing process, focused on carbonatation step, enabled to highlight points which could be enhanced to optimize the extraction yield and the quality of extracted alginates. Optimization of extraction parameters for recovery of alpha-amylase from the fermented bran of Bacillus circulans GRS313 BABT

The extraction of alpha-amylase in the solid state fermentation of wheat bran by Bacillus circulans GRS313 was optimized. Among various solvents tested, maximum extraction was achieved when 2.5% (v/v) glycerol was added. The optimum conditions for extraction were 2.5 hrs soaking time at 30° C under recirculation and agitated condition, which yielded 590 U/g bran of enzyme activity. Whereas under stationary soaking condition the activity of the enzyme was 439.58U/g bran. With repeated wash under the above optimum conditions showed that 600 U/g and 184.2 U/g of enzyme extracted in the 1st and 2nd washes respectively and only 36.84 U/g was recovered in the 3rd wash

Potencial osmótico y absorción nutrimental en el cultivo in vitro de Agave potatorum Zucc. Colegio de Postgraduados

Con el objeto de conocer el efecto del potencial osmótico de las sales inorgánicas del medio de Murashige y Skoog se determinó el contenido de humedad, peso de materia seca, concentración y extracción nutrimental en brotes de Agave potatorum Zucc. Se establecieron medios de cultivo con los siguientes potenciales osmóticos: -0.187, -0.139, -0.094 y -0.050 MPa. El experimento se estableció con un diseño completamente al azar con tres repeticiones. Los datos obtenidos se sometieron a un análisis de varianza y comparación de medias. Después de 90 días del establecimiento del experimento se encontraron diferencias estadísticas significativas entre tratamientos para las variables evaluadas. Purification and some properties of pectinesterase from potato (Solanum tuberosum L.) alpha cultivar BABT

Pectinesterase was extracted from potato alpha cultivar, purified and partially characterized The used protocol resulted in a 58.8-fold purification (51 850.2 units/mg protein) with 15.5% recovery of pectinesterase activity. The purified enzyme had a molecular weight of 27 kDa and its isoelectric point was around 4.5 with pH and temperature optima of 8.0 and 60°C, respectively. The purified enzyme had a single symmetric peak of specific activity after chromatographic steps. The homogeneity of the purified pectinesterase was confirmed by gel filtration and polyacrylamide electrophoresis gel.

The objective of this study was to optimize the extraction of chitin by acid proteolysis. The novelty of the method is based on the stabilization of pH by the balance between substrate composition and the acid solvent. This principle allows a simultaneous demineralization and deproteinization, the two main reactions associated with the purification of chitin. To evaluate the performance of this purification, the composition of the substrate and products was characterized. Different methods of quantification of chitin and proteins have been compared. Apesar da qualidade elevada, o processo de extração do óleo presente na polpa da bacaba é realizado na região Norte prioritariamente na forma artesanal, sendo os frutos aquecidos em água para que ocorra o amolecimento da polpa. Após essa etapa, realiza-se maceração manual e novo aquecimento com água para separação do óleo sobrenadante. Essa exposição a elevadas temperaturas prejudica a qualidade final do óleo, além de ser um método demorado e de baixo rendimento. Em contraposição, extrações químicas resultam em maior rendimento, contudo podem deixar resíduos nos óleos que não são adequados para a produção de comestíveis. Study on the microalgal pigments extraction process: Performance of microwave assisted extraction ArchiMer

The performance of microwaves irradiation (MAE and VMAE) to extract pigments from two marine microalgae was compared to conventional processes (cold and hot soaking and ultrasound-assisted extraction). Pigments were quantified by RP-HPLC and extraction performance was assessed regarding rapidity, reproducibility and extraction yields. Scanning electron microscopy was used at all extraction steps to assess the impact of the process on microalgal cell integrity. Freeze-drying and pigments extraction preserved microalgae cell integrity (except sonication) and evoked agglutination in superposed cells layers. Unlike other resources such as petroleum, coal, and copper, forests are renewable. Yet, in many respects forests historically have been treated as a nonrenewable resource in that forest stocks were depleted or "mined" and loggers moved on to exploit other "deposits." The lands were often put to other uses, typically agricultural, or allowed to regenerate naturally. This paper looks at technical change in forest extraction, i.e., logging under a number of different conditions. It finds that, on average, labor productivity has been increasing in recent decades. Physico-chemical, biological and functional properties of proteins extracted from organic rice bran with different methods were determined. Rice bran protein (RBP) obtained from enzymatic (phytase-xylanase) extraction showed the lowest color difference (ΔE, P<0.05). Bulk density of RBP was ranged between 0.71-0.73 g/ml (P>0.05). Total soluble solid and pH of RBP obtained from different extraction methods were not significantly different (P>0.05). Sonication method provided the extract with the highest total phenolic content (3.01 mg tannic acid/g RBP), DPPH radical scavenging activity (62.29%), and ferric reducing activity power (FRAP 2.00 mM).
